Major research interests:

1. Ethylene receptor signaling and plant growth and stress responses

Ethylene plays important roles in many aspects of plant growth and development. It is also involved in stress response. However, how ethylene signaling affects plant abiotic stress response remains unclear. In recent years, we have been focusing on the role of this signaling pathway in salt stress responses. Two tobacco ethylene receptor genes NTHK1 and NTHK2 have been cloned and characterized. These two subfamily II receptors have Ser/Thr kinase activity, unlike the Arabidopsis subfamily I receptor ETR1 that has His kinase activity. The Ser/Thr kinase activity may have specific roles in signaling. Overexpression of NTHK1 confers salt sensitivity and ACC can suppress this sensitivity, indicating that ethylene signaling is required for salt tolerance. This result is also consistent with the negative regulation between ethylene and its receptors. Ethylene signaling-regulated genes and a number of stress-regulated transcription factor and effector genes have been studied for their roles in plant abiotic stress response. Further study on the roles of kinase activity in receptor signaling and identification of NTHK1-interacting proteins should reveal the mechanisms of the receptor signaling. Ethylene receptors from monocots rice plants were also identified and their biochemical properties and functions in rice were investigated. Rice ethylene response mutants were also identified and characterized. These studies should disclose the features of ethylene signaling in monocot plants.

2. Functional analysis of regulatory genes in regulation of oil biosynthesis and abiotic stress response in soybean plants.

Soybean is an important crop for both oil and protein worldwide. The oil biosynthesis pathway has been established. However, the regulatory mechanism for this pathway at the transcriptional level is largely unknown. Using bioinformation, systems biology, genomic and molecular approaches, we have identified genes involved in control of lipid biosynthesis. These included Dof-type transcription factors and other-related genes. Engineering of these genes may potentially improve plant oil content. Abiotic stress responsive genes were also identified and their roles in abiotic stress tolerance were investigated in transgenic Arabidopsis and/or Lotus plants. A number of transcription factor genes and other genes have been found to incease stress tolerance. These genes will be used to improve stress tolerance in legume or other relevant plants.
